The drugs used to treat amyloid-β (Aβ) oligomers and tau-associated neurodegenerative disorders often lead to cytotoxicity and neurotoxicity. Silymarin (SM), a flavonoid extracted from Milk Thistle plant (Silybum marianum), has been reported to possess antioxidant properties. The central nervous system (CNS) is in continuous exposure to oxidative insults during its lifespan. Here, the effect of aqueous extract of SM in aged rat brainstem superficial white matter on antioxidant status and its possible molecular mechanisms is evaluated. Moreover, the effect of its co-administration with Aβ-42 is also investigated. The antioxidative efficiency of SM is assessed using the reactive oxygen species (ROS) generation and antioxidant capacity assay of brainstem superficial white matter. The amount of malondialdehyde (MDA), nitrite, and superoxide dismutase (SOD) activity was measured as a marker of oxidative damages. In addition, the differential expression of antioxidant genes and proteins is investigated in this study. The result obtained showed that brainstem superficial white matter is subjected to oxidative stresses in aged rats. It also manifested that SM inhibited Aβ-42-induced ROS generation and restored the antioxidant capacity of aged rats in brainstem superficial white matter. The study proposes that the potential mechanism of SM is based on its antioxidant capacity by enhancing the expression of antioxidant genes and proteins. The possible mechanism of SM as an effective antioxidant for management of neurodegenerative disorders is discussed, and it could provide a novel therapeutic approach to the management of such diseases.Adenosine A3 receptors in human coronary artery vasculature regulate vasodilatory function.
